Not really, plus you risk damaging the valve diaphragm, but it's probably going to get covered anyway, and will need replaced at some point. I have used Polymarine's sealant on a slow leak on a Zodiac years ago, so the bottle simply gets poured in, inflate tube, rotate the boat (easy in a SIB), then inflate to pressure. I think what you're describing will work so you can get the internal tube surface covered.
I'd have thought you'd be better off trying to identify the leak, as these products provide nothing more than a thin membrane, so long term - not sure how it stands up?
If you think the pin hole may be in a hard to access area, try filling the boat with 2-3 inches of soapy water and use the trailer to move the water back and forth and see if that reveals anything. Work you're way around the boat with a spray bottle, section at a time.
